Background of Solubilizer Excipients

Solubilizer refers to a surfactant with solubilizing ability. Solubilization refers to the process in which poorly soluble drugs increase their solubility in a solvent and form a solution under the action of surfactants. The properties of the solubilizer, the nature of the solubilizer, the HLB value of the solubilizer, temperature, and the amount of the solubilizer are all influencing factors for the solubilization effect. In the presence of colloidal surfactant particles, the process of increasing the solubility of poorly soluble drugs and forming a clear solution is called solubilization. Commonly used solubilizers include polysorbates and polyoxyethylene fatty acid esters.

Types of Solubilizer Excipients:

There are two main types of surfactants currently used as solubilizers in pharmaceutical preparations: one is anionic surfactants, which are mainly used in external preparations; the other is nonionic surfactants, which are the most widely used.

Anionic

The part of anionic solubilizer that plays a surface active role is anion. Because of its strong physiological effects, such as hemolysis, irritation, etc., it is generally used in external preparations.

Non-ionic

The non-ionic solubilizer does not dissociate in water. The hydrophilic group in the molecule is composed of polyols and their polymers, and the lipophilic group is composed of various higher fatty acids or higher fatty alcohols and alkyl or aryl groups. There are many different types of ester bonds or ketone bonds combined with hydrophilic groups. Due to its chemical non-dissociation, this type of solubilizer is not easily affected by electrolytes and pH value of the solution, has less toxicity and hemolysis, and is currently the most widely used. It can be used for oral, external, injection and other dosage formulations.

Generally, cationic surfactants are not used as solubilizers. Anionic surfactants are only used for external preparations, while nonionic surfactants are widely used. They are used in oral preparations, external preparations and even injections.

Solubilizers are often used in the preparation of liquid preparations and injections. Only non-toxic surfactants can be used as solubilizers for internal preparations and injections. External preparations also need to consider their irritation.
