Customization and Modification for Dendrimers

Dendrimers are capable of encapsulating drugs through electrostatic, van der Waals and H-bonding forces or chemical bonds due to their unique properties (e.g., well-defined size, spherical shape and internal void space). The dendrimer consists of three parts: the central core of the molecule, the inner layer of the branched unit, and the outer layer of the functional group. Common types include poly-lysine (PLL), poly-propyleneimine (PPI), poly-ethyleneimine (PEI), poly-aryl ethers, and poly-amidomethylamine ( PAMAM). However, the toxicity of dendrimers is the biggest obstacle limiting their clinical applications. CD Formulation typically uses polyethylene glycol (PEG) and its derivatives for surface modification. PEG and its derivatives can improve their water solubility, reduce enzymatic cleavage by metabolic enzymes and immune system in vivo, and prolong the retention time of drugs in vivo. Currently, we can successfully modify dendrimers, including bromination, amination, carboxymethylation, toluenesulfonation, methanesulfonation, aldolization, or methoxypolyethylene glycol (MPEG) chemistry. Dendrimers that are anionic or polyethylene glycolized and their derivatives are significantly less cytotoxic compared to cationic dendrimers with other surface groups.
