Extracellular vesicles (EVs) are a class of submicron lipid membrane vesicles secreted by cells with a bilayer lipid membrane structure. Depending on the formation process and cellular origin, extracellular vesicles (EVs) include microvesicles (MVs), exosomes, and apoptotic vesicles. Microvesicles, generally 50 nm to 1 μm in diameter, are nanoscale membrane-bound vesicles secreted by cells directly through the plasma membrane outgrowth vesicles, which are subcellular physiological carriers present in all body fluids.

Advantages of Extracellular Vesicles as Drug Delivery Vehicles

  • EVs contain a lipid layer with good mobility and a corresponding membrane protein on the lipid layer, which is strongly targeted.
  • EVs are modified to enhance their fusion with membranes and uptake by cells, and are easily acquired by target cells actively or passively, thus improving drug delivery efficiency.
  • The composition of EVs is very similar to that of human own cells, which is non-immunogenic and has good biocompatibility.
  • EVs are self-cellular products that prevent their own phagocytosis by macrophages, degradation by endosomal pathways and lysosomes, and thus have long circulation characteristics and can deliver the transported drugs directly to the target cells.
